Use | used in the manufacture of pharmaceuticals, pesticides, veterinary drugs, dyes, etc. Sulfonamides are intermediates in the production of famotidine, a gastric drug. |
production method | in a 3.9 ml three-mouth bottle with reflux condenser and stirrer, add 0.12g (mol) of sodium hydroxide, add a small amount of water as solvent, stir, heat to fully dissolve, add 23.3g(0.12mol) of 2-chloro-n, N-dimethylnicotinamide, then reflux for 3H to obtain N,N-dimethylnicotinyl polysulfide sodium salt solution. Add water to the above reaction solution, and cool to 50oC; Add hydrochloric acid to adjust PH equal to 1, and react for 1H, then filter while hot, filter residue is rinsed with hot water, the filtrates were combined to give an aqueous solution of 2-mercapto-n, N-dimethylnicotinamide to be used. Add the above-mentioned 2-mercapto-n, N-dimethylnicotinamide aqueous solution to a 250-ml triplicate bottle with a chlorine tube and a tail gas receiving device and with mechanical stirring, and cool to room temperature, chlorine gas 25.6g(0.36ml) was passed and reacted for 0.5h at room temperature. The solvent 1, 2-dichloroethane was then added to extract twice, and the oil layers were combined, thereby obtaining a solution of 2-chlorosulfonyl-n, N-dimethylnicotinamide in dichloroethane to be used. Add the above-mentioned solution of 2-chlorosulfonyl-n, N-dimethylnicotinamide in dichloroethane to a 500ml three-mouth bottle with ammonia tube and tail gas receiver and with mechanical stirring, ammonia gas is passed at room temperature until PH is greater than 9, and then the reaction is continued to stir at room temperature for 15min, then 26.1g of white powder is obtained by filtration, and 19.7g of white crystal is obtained by recrystallization, with content of 91.0%, the overall yield was 65.1% based on 2-chlorosulfonyl-n, N-dimethylnicotinamide. |
